The story takes place after 5 years of Super Mario Odyssey. Mario and Cappy were invited to a party with Princess Peach and Tiara, and invite their friends: Luigi and Helmer, Wario and Bowler, Waluigi and Fedoro, Toad and Fedo, Toadette and Sunny, Baby Mario and Baby Cappy, Baby Luigi and Baby Helmer, Baby Wario and Baby Bowler, and Baby Waluigi and Baby Fedoro. They went into the Odyssey, but Bowser and Toppy command their minions to capture them, leaving the only hero left, Yoshi, sleeping on the roof of Peach's castle.
Meanwhile in the Cap Kingdom, a ghost arrives in the Odyssey and wakes Yoshi up, and then Yoshi catches up with him. The ghost introduces himself as Hatson then tells Yoshi that Mario and his friends have not went to the party yet and Princess Peach and Tiara have got kidnapped. Hatson then turns into Yoshi's tongue and teams up with him, then they collect Power Moons and save their friends. Then the eleven heroes collect the rest of the Power Moons in order to save Princess Peach and Tiara from Bowser and Toppy.

Yoshi
Yoshi is the first and main playable character. He can use his tongue to each enemies and items, can flutter jump, jumps high, and has good speed, but is not as powerful as the others.

Mario is the first character rescued. He has balanced stats and can wall jump. He is required.
Defeat Goomboss as Yoshi
Luigi
Luigi is the second character rescued. He has high jumps, but low traction. He is optional.
Defeat King Boo as Mario
Wario
Wario is the third character rescued. He is strong, but has low jumps and speed. Like Luigi, he is optional.
Defeat Chief Chilly as Luigi
Waluigi
Waluigi is the fourth character to be rescued. He has good swimming capibilities but low power, Like Luigi and Wario, he is optional.
Defeat Petey Piranha as Wario
Toad
Toad is the fifth character rescued. He is speedy, but is weak and has low jumps. Like Luigi, Wario and Waluigi, he is optional.
Defeat Wingo as Waluigi
Toadette
Toadette is the sixth character rescued. She can hover shortly after jumping but is weak and slow. Like Luigi, Wario, Waluigi and Toad, she is optional.

Baby Mario
Baby Mario is the seventh character to be rescued. He is fast but has bad jumps. Like Luigi, Wario, Waluigi, Toad and Toadette, he is optional.

Baby Luigi is the eighth character to be rescued. He is fast but has low traction. Like Luigi, Wario, Waluigi, Toad, Toadette and Baby Mario, he is optional.
